Optimized preparation of -polyglutamic acid/chitosan nanocapsule

This paper described the preparation of novel biodegradable nanocapsule based on self-assembly of -Polyglutamic acid (-PGA) and chitosan (CS). After the Plackett-Burman design (PBD), the impact of mass concentration and volume of -PGA and pH value of CS were characterized by size and PDI of the nanocapsule. A Box-Behnken design (BBD) was used to optimize the preparation of the nanocapsule. The optimized condition was: pH value of CS was 4.0; volume of γ-PGA was 18mL; mass concentration of γ-PGA was 0.1g/L. The Z-Ave and PDI of the nanocapsules prepared under the best conditions were 175 nm and 0.15 respectively. In this work, we have shown that nano-sized particles have been successfully assembled from the γ-PGA and chitosan without employing covalent linkages between these biopolymers. These results will provide a novel concept in the design of carrier systems composed of polyion complex (PIC).
